A hierarchical Bayesian approach to ranking and selection as well as estimation of related means in two—way models is considered. Using the method of Monte Carlo simulation with importance sampling, we are able to carry out efficiently the three or four dimensional integrations as needed. Children represent a large underserved population of “therapeutic orphans,” as an estimated 80% of children are treated off‐label. However, pediatric drug development often faces substantial challenges, including economic, logistical, technical, and ethical barriers, among others. Among many efforts trying to remove these barriers, increased recent attention has been paid to extrapolation; that is, the leveraging of available data from adults or older age groups to draw conclusions for the pediatric population. The Bayesian statistical paradigm is natural in this setting, as it permits the combining (or “borrowing”) of information across disparate sources, such as the adult and pediatric data. In this paper, authored by the pediatric subteam of the Drug Information Association Bayesian Scientific Working Group and Adaptive Design Working Group, we develop, illustrate, and provide suggestions on Bayesian statistical methods that could be used to design improved pediatric development programs that use all available information in the most efficient manner. A variety of relevant Bayesian approaches are described, several of which are illustrated through 2 case studies: extrapolating adult efficacy data to expand the labeling for Remicade to include pediatric ulcerative colitis and extrapolating adult exposure‐response information for antiepileptic drugs to pediatrics.  相似文献

We display the first two moment functions of the Logitnormal(μ, σ2) family of distributions, conveniently described in terms of the Normal mean, μ, and the Normal signal-to-noise ratio, μ/σ, parameters that generate the family. Long neglected on account of the numerical integrations required to compute them, awareness of these moment functions should aid the sensible interpretation of logistic regression statistics and the specification of “diffuse” prior distributions in hierarchical models, which can be deceiving. We also use numerical integration to compare the correlation between bivariate Logitnormal variables with the correlation between the bivariate Normal variables from which they are transformed.  相似文献

This paper considers regression models for cross‐section data that exhibit cross‐section dependence due to common shocks, such as macroeconomic shocks. The paper analyzes the properties of least squares (LS) estimators in this context. The results of the paper allow for any form of cross‐section dependence and heterogeneity across population units. The probability limits of the LS estimators are determined, and necessary and sufficient conditions are given for consistency. The asymptotic distributions of the estimators are found to be mixed normal after recentering and scaling. The t, Wald, and F statistics are found to have asymptotic standard normal, χ2, and scaled χ2 distributions, respectively, under the null hypothesis when the conditions required for consistency of the parameter under test hold. However, the absolute values of t, Wald, and F statistics are found to diverge to infinity under the null hypothesis when these conditions fail. Confidence intervals exhibit similarly dichotomous behavior. Hence, common shocks are found to be innocuous in some circumstances, but quite problematic in others. Models with factor structures for errors and regressors are considered. Using the general results, conditions are determined under which consistency of the LS estimators holds and fails in models with factor structures. The results are extended to cover heterogeneous and functional factor structures in which common factors have different impacts on different population units.  相似文献

Summary This expository paper provides a framework for analysing de Finetti's representation theorem for exchangeable finitely additive probabilities. Such an analysis is justified by reasoning of statistical nature, since it is shown that the abandonment of the axiom of σ-additivity has some noteworthy consequences on the common interpretation of the Bayesian paradigm. The usual (strong) fromulation of de Finetti's theorem is deduced from the finitely additive (weak) formulation, and it is used to solve the problem of stating the existence of a stochastic process, with given finite-dimensional probability distributions, whose sample paths are probability distributions. It is of importance, in particular, to specify prior distributions for nonparametric inferential problems in a Bayesian setting. It is desired to select that distribution with the largest parameter value unless this value is smaller than some fixed standard θ0 in which case no distribution is to be selected. ABSTRACT

Given a sample from a finite population, we provide a nonparametric Bayesian prediction interval for a finite population mean when a standard normal assumption may be tenuous. We will do so using a Dirichlet process (DP), a nonparametric Bayesian procedure which is currently receiving much attention. An asymptotic Bayesian prediction interval is well known but it does not incorporate all the features of the DP. We show how to compute the exact prediction interval under the full Bayesian DP model. However, under the DP, when the population size is much larger than the sample size, the computational task becomes expensive. Therefore, for simplicity one might still want to consider useful and accurate approximations to the prediction interval. For this purpose, we provide a Bayesian procedure which approximates the distribution using the exchangeability property (correlation) of the DP together with normality. We compare the exact interval and our approximate interval with three standard intervals, namely the design-based interval under simple random sampling, an empirical Bayes interval and a moment-based interval which uses the mean and variance under the DP. However, these latter three intervals do not fully utilize the posterior distribution of the finite population mean under the DP. Using several numerical examples and a simulation study we show that our approximate Bayesian interval is a good competitor to the exact Bayesian interval for different combinations of sample sizes and population sizes.  相似文献

Summary.  We discuss the analysis of data from single-nucleotide polymorphism arrays comparing tumour and normal tissues. The data consist of sequences of indicators for loss of heterozygosity (LOH) and involve three nested levels of repetition: chromosomes for a given patient, regions within chromosomes and single-nucleotide polymorphisms nested within regions. We propose to analyse these data by using a semiparametric model for multilevel repeated binary data. At the top level of the hierarchy we assume a sampling model for the observed binary LOH sequences that arises from a partial exchangeability argument. This implies a mixture of Markov chains model. The mixture is defined with respect to the Markov transition probabilities. We assume a non-parametric prior for the random-mixing measure. The resulting model takes the form of a semiparametric random-effects model with the matrix of transition probabilities being the random effects. The model includes appropriate dependence assumptions for the two remaining levels of the hierarchy, i.e. for regions within chromosomes and for chromosomes within patient. We use the model to identify regions of increased LOH in a data set coming from a study of treatment-related leukaemia in children with an initial cancer diagnostic. The model successfully identifies the desired regions and performs well compared with other available alternatives.  相似文献
