The quantification of the relationship between the amount of microbial organisms ingested and a specific outcome such as infection, illness, or mortality is a key aspect of quantitative risk assessment. A main problem in determining such dose-response models is the availability of appropriate data. Human feeding trials have been criticized because only young healthy volunteers are selected to participate and low doses, as often occurring in real life, are typically not considered. Epidemiological outbreak data are considered to be more valuable, but are more subject to data uncertainty. In this article, we model the dose-illness relationship based on data of 20 Salmonella outbreaks, as discussed by the World Health Organization. In particular, we model the dose-illness relationship using generalized linear mixed models and fractional polynomials of dose. The fractional polynomial models are modified to satisfy the properties of different types of dose-illness models as proposed by Teunis et al . Within these models, differences in host susceptibility (susceptible versus normal population) are modeled as fixed effects whereas differences in serovar type and food matrix are modeled as random effects. In addition, two bootstrap procedures are presented. A first procedure accounts for stochastic variability whereas a second procedure accounts for both stochastic variability and data uncertainty. The analyses indicate that the susceptible population has a higher probability of illness at low dose levels when the combination pathogen-food matrix is extremely virulent and at high dose levels when the combination is less virulent. Furthermore, the analyses suggest that immunity exists in the normal population but not in the susceptible population.  相似文献

The combined model accounts for different forms of extra-variability and has traditionally been applied in the likelihood framework, or in the Bayesian setting via Markov chain Monte Carlo. In this article, integrated nested Laplace approximation is investigated as an alternative estimation method for the combined model for count data, and compared with the former estimation techniques. Longitudinal, spatial, and multi-hierarchical data scenarios are investigated in three case studies as well as a simulation study. As a conclusion, integrated nested Laplace approximation provides fast and precise estimation, while avoiding convergence problems often seen when using Markov chain Monte Carlo.  相似文献

We develop local influence diagnostics to detect influential subjects when generalized linear mixed models are fitted to incomplete longitudinal overdispersed count data. The focus is on the influence stemming from the dropout model specification. In particular, the effect of small perturbations around an MAR specification are examined. The method is applied to data from a longitudinal clinical trial in epileptic patients. The effect on models allowing for overdispersion is contrasted with that on models that do not.  相似文献

For the linear mixed model the approach leads to easily interpretable and computationally convenient expressions, not only highlighting influential subjects, but also which aspect of their profile leads to undue influence on the model's fit [17 E. Given the model's nonlinear structure, these authors did not derive interpretable components but rather focused on a graphical depiction of influence. In this paper, we consider GLMMs for binary, count, and time-to-event data, with the additional feature of accommodating overdispersion whenever necessary. For each situation, three approaches are considered, based on: (1) purely numerical derivations; (2) using a closed-form expression of the marginal likelihood function; and (3) using an integral representation of this likelihood. Unlike when case deletion is used, this leads to interpretable components, allowing not only to identify influential subjects, but also to study the cause thereof. The methodology is illustrated in case studies that range over the three data types mentioned.  相似文献

This paper will highlight to what extend Eisenstadt’s theory connects to the approach of historic generations. At first, his early comparative work “From Generation to Generation” is reconstructed. In this book, Eisenstadt presents a model of generations based on structural-functionalist assumptions; he conceptualizes the relation between generations and social change as a problem of system integration of specific age-groups on the one hand and the regulation of tensions between these groups on the other hand. Using the youth- and student movements as an example, this article shows how Eisenstadt’s interpretations change with the development of his theory. In the second part of this paper, it will be proposed how his approach can be developed further by interlocking his concept more closely with Mannheim’s theory of generations as well as other recent findings of socialization theory. It will furthermore be discussed, how his modernization theory has the potential to solve a theoretical problem inherent in Mannheim’s approach.  相似文献

Based on sero-prevalence data of rubella, mumps in the UK and varicella in Belgium, we show how the force of infection, the age-specific rate at which susceptible individuals contract infection, can be estimated using generalized linear mixed models (McCulloch & Searle, 2001). Modelling the dependency of the force of infection on age by penalized splines, which involve fixed and random effects, allows us to use generalized linear mixed models techniques to estimate both the cumulative probability of being infected before a given age and the force of infection. Moreover, these models permit an automatic selection of the smoothing parameter. The smoothness of the estimated force of infection can be influenced by the number of knots and the degree of the penalized spline used. To determine these, a different number of knots and different degrees are used and the results are compared to establish this sensitivity. Simulations with a different number of knots and polynomial spline bases of different degrees suggest - for estimating the force of infection from serological data - the use of a quadratic penalized spline based on about 10 knots.  相似文献

This paper examines the impact of new technology on the labour process and labour market position of clerical and administrative workers in The Federal Republic of Germany. Data are drawn from several recent major case studies of such workers by German research teams. The paper's contributions to the ‘Labour Process’ debate are twofold. Firstly, it examines the position of white-collar workers to show that, contrary to Braverman, their work situation differs crucially in several respects from that of manual workers, despite some convergence in recent decades. Secondly, it shows that variations in the organization of the labour process between capitalist societies are much wider than is commonly recognized. The paper argues that historically evolved social structural and cultural features shape the position of German white-collar workers in the labour process and distinguish it in several respects from that of comparable groups in Anglo-Saxon countries.  相似文献

This paper deals with the analysis of data from a HET‐CAMVT experiment. From a statistical perspective, such data yield many challenges. First of all, the data are typically time‐to‐event like data, which are at the same time interval censored and right truncated. In addition, one has to cope with overdispersion as well as clustering. Traditional analysis approaches ignore overdispersion and clustering and summarize the data into a continuous score that can be analysed using simple linear models. In this paper, a novel combined frailty model is developed that simultaneously captures all of the aforementioned statistical challenges posed by the data.
