钠氢交换体1 mRNA在心衰患者心肌组织中的表达

摘    要:目的观察钠氢交换体1(sodium—hydrogen exchanger1,NHE1)在心力衰竭患者心肌组织中的表达,探讨NHE1在心衰发生和发展中的作用。方法采用实时荧光定量聚合酶链反应(FQ—PCR)检测35例心力衰竭患者心肌组织NHE1mRNA的表达水平。结果实时荧光定量PCR(FQ—PCR)检测心功能Ⅰ级组、心功能Ⅱ级组、心功能Ⅲ级组心力衰竭患者心肌组织NHE1mRNA的△Ct值分别为6.29±0.66、5.01±0.60、4.40±0.74。与心功能Ⅰ级组比较,在心功能Ⅱ级组、心功能Ⅲ级组患者心肌组织中NHE1mRNA表达明显高于心功能Ⅰ级组(P〈0.01),同时与心功能Ⅱ级组比较,心功能Ⅲ级组患者心肌组织中NHE1mRNA水平增高,且差异有统计学意义(P〈0.05)。结论心衰患者心肌组织中存在NHE1mRNA的高水平表达,提示NHE1可能在心衰的发生和发展过程中起着重要作用。

Abstract:Objectives To detect the Expreesion of Myocardial Sodium-Hydrogen Exchanger 1 mRNA in heart failure, and to study the effects of NHE1 in the incidence of heart failure. Methods The Expreesion of Myocardial Sodium-Hydrogen Exchanger 1 mRNA in 35 heart failure patients was detected by using real-time fluorescence quantitative PCR (FQ-PCR). Results With the real-time fluorescence quantitative PCR method,the ACt value of class Ⅰ group,class Ⅱ group,class Ⅲgroup were 6.29±0.66,5.01±0.60,4.40±0.74,the expresion of Myocardial Sodium-Hydrogen Exchanger 1 mRNA in group of class Ⅱand class Ⅲ were significantly higher than class Ⅰ group (P〈0.01). At the same time,Comparison with the group of class Ⅱ ,the Expreesion of Myocardial Sodium-Hydrogen Exchanger 1 mRNA in group of class m was higher level,and the difference was statistically significant (P〈0.05). Conclusion The mRNA Expreesion of gene Sodium-Hydrogen Exchanger 1 (NHE1) was up-regulated in heart failure,that may prompt the occurrence of heart failure in the process of development and plays an important role.
