植物细胞凋亡的诱导及检测

摘    要:本研究以洋葱鳞茎内表皮细胞及大蒜根尖生长点细胞为材料,分别用不同浓度的离子胁迫及过氧化氢诱导处理,观察植物细胞凋亡过程中的形态变化.结果显示:O.4mol/L的Nacl和CaCl2处理2小时即可诱导内表皮细胞出现明显的细胞质皱缩,染色质凝聚等凋亡现象;1%H2O2处理4小时可诱导生长点细胞出现分裂后期染色体桥及间期细胞核膨胀,形成空泡,出现染色质异固缩化等典型的凋亡特征.此研究为细胞凋亡实验教学提供了经济、快捷、有效的诱导和检测方法.

The Inducement and Detection of Apoptosis in Plant Cells

Abstract:In this paper, endocutiele cells of onion squamous bulb and the growing point cells of the garlic root tip were used as the material to detect the morphological changes of the apoptotic plants cells induced by ionic stress ( NaCl, CaCl2 ) and hydrogen peroxide. We found that 0.4 moVL ionic stress ( NaCl, CaCl2 ) can induce distinct shrinkage of cytoplasm and concentration of chromatin in eudocuticle cells only within two hours. The growing point cells treated with 1% H2 O: for 4 hours can induce a series of typically apoptotic characteristics in unclei, such as Chromosome Bridge in anaphase, expansion nuclei with vacuole and heteropyknosis of chromatin, etc. This study provided an economical, speedy and effective method on induction and detection for the apoptosis experimental teaching.
