长春西汀渗透泵控释片在Beagle犬体内的药动学研究

摘    要:考察长春西汀(VIN)渗透泵控释片在Beagle犬体内的药动学特点.采用高效液相法,以羟孕酮为内标,测定血浆中VIN的血药浓度,使用3P97软件处理计算主要药动学参数.VIN渗透泵片和普通片血浆中VIN的t1/2分别为(1.97±0.34)和(0.85±0.23)h;Cmax分别为(15.78±2.98)和(80.25±12.38)ng·mL-1;Tmax分别为(3.58±0.55)和(0.76±0.23)h.VIN的相对生物利用度分别为(96.11±24.14)%.VIN渗透泵片血药浓度平稳,可较长时间保持血药浓度.

Abstract:To investigate the pharmacokinetics and bioavailability of VIN osmotic pump tablet in Beagle dog.Medroxyprogesterone was used as internal standard for VIN drug and the concentrations were detected by HPLC;The concentration-time data were fitted using 3P97 pharmacokinetics program.The main pharmacokinetics parameters of VIN of two preparations were as follows:t1/2(1.97±0.34)and(0.85±0.23)h;Cmax(15.78±2.98)and(80.25±12.38)ng·mL-1;Tmax(3.58±0.55)and(0.76±0.23)h,respectively.The relative bioavailability of VIN was(96.11±24.14)%.Plasma concentration of VIN osmotic pump tablets are constant and effective plasma drug concentration is kept for a long time.
