Urine-concentrating ability is decreased in the aging mammalian kidney. Studies have revealed various changes in kidney function that occur with aging and may explain the reduced ability to concentrate urine. Recently, the genes encoding many of the water- and solute-transport proteins and the vasopressin receptor, all of which are involved in urine concentration, have been cloned. Therefore, the molecular mechanisms that cause the reduction in urine-concentrating ability with aging can now be deciphered. In this Perspective, I discuss recent experiments designed to characterize this change in kidney function in aging mammals.  相似文献

Erectile dysfunction (ED) is one of the most common disorders in male and is often associated with other age-related comorbidities. The aging process affects the structural organization and function of penile erectile components such as smooth muscle cell and vascular architecture. These modifications affect penile hemodynamics by impairing cavernosal smooth muscle cell relaxation, reducing penile elasticity, compliance and promoting fibrosis. This review aims to identify the mechanisms of ED in the penile aging process in experimental and clinical data. It also highlights areas that are in need of more research. The search strategies yielded total records screened from PubMed. Clarification of the molecular mechanisms that accompanies corpus cavernosum aging and aging-associated ED will aid new perspectives in the development of novel mechanism-based therapeutic approaches. Age is not a limiting factor for ED medical management, and it is never too late to treat. Hypogonadism should be managed regardless of age, and synergistic effects have been found during testosterone (T) replacement therapy when used along with oral phosphodiesterase-5 (PDE-5) inhibitors. Therefore, the clinical management of ED related to aging can be done by therapeutic interventions that include PDE-5 inhibitors, and other pharmacological treatments.  相似文献

The budding yeast Saccharomyces cerevisiae has long served as a model organism for the study of basic cellular processes. Its short generation time, well-established molecular genetics, and fully sequenced genome have made this organism a favorite of researchers in diverse fields. Much of the information obtained has been shown to apply to higher eukaryotes, including humans. Recently, researchers have begun using yeast to tackle one of the outstanding questions in science: How and why do organisms age? The identification of individual genes in yeast that can affect the aging process itself has elevated this single-celled fungus to full contender status in the aging field. In this Perspective, we present two fundamentally different measures of aging in yeast: replicative life-span and stationary phase survival (chronological life-span). We describe the benefits and limitations of each and present models that attempt to explain these "aging" phenomena. Finally, we present compelling evidence that the use of yeast as a model system will ultimately prove beneficial to the study of human aging.  相似文献

Although several biomarkers of aging have been described in the literature, it is only recently that gerontologists have started to search for molecular biomarkers of aging. A gene or a set of genes that are expressed in a wide range of tissues and exhibit an age-dependent, easily quantifiable increase in their expression represent a possible molecular biomarker of aging. Because the physiology of an organism is profoundly affected by the pattern of gene expression, it is hoped that molecular biomarkers of aging will more accurately predict the physiological age of an organism than the chronological age. A recent report from Sharpless's laboratory examines the possibility that the tumor suppressors p16 and ARF (encoded by the INK4a/ARF locus) represent molecular biomarkers of aging in rodent models.  相似文献

Evaluation of physical functioning is a key issue in clinical geriatrics and in aging research. In recent years, different physical performance batteries in which individuals are asked to perform several tasks and are evaluated using different criteria have been designed and used in elderly populations. These batteries include different types of test which range from basic motor abilities to relevant everyday activities, depending on the construct area in the domain of physical function that must be measured. This paper reviews and classifies the main physical functioning batteries that can be found in the scientific field of aging research in order to provide knowledge on selection, administration, and interpretation of this indispensable assessment tools.  相似文献

Aging is a complex process that involves the gradual functional decline of many different tissues and cells. Gene expression microarray analysis provides a comprehensive view of the gene expression signature associated with age and is particularly valuable for understanding the molecular mechanisms that contribute to the aging process. However, because of the stochastic nature of the aging process, animals of the same chronological age often manifest great physiological differences. Therefore, profiling the gene expression pattern of a large population of aging animals risks either exaggerating or masking the changes in gene expression that correspond to physiological aging. In a recent paper, Golden and Melov surveyed the gene expression profiles of individual aging Caenorhabditis elegans, hoping to circumvent the problem of variability among worms of the same chronological age. This initial analysis of age-dependent gene expression in individual aging worms is an important step toward deciphering the molecular basis of physiological aging.  相似文献

The countries of the Middle East all have traditional social insurance type social security programs. The move towards defined contribution individual accounts that is occurring in some regions has not affected this region The social security programs in the Middle East are not facing the problems of financing found in Europe and North America, in part because they still have relatively high fertility rates and are thus little affected by population aging. They tend to have low retirement ages and some of the wealthy countries of the region provide very generous benefits. Many of them need to consider reforms that raise retirement ages. A characteristic of many of the social security old-age benefit programs of the region is that they exclude foreign workers, who in some countries account for more than half the workforce.  相似文献

In this work, we study graph clustering problems associated with structural balance. One of these problems is known in computer science literature as the correlation-clustering (CC) problem and another (RCC) can be viewed as its relaxed version. The solution of CC and RCC problems has been previously used in the literature as tools for the evaluation of structural balance in a social network. Our aim is to solve these problems to optimality. We describe integer linear programming formulations for these problems which includes the first mathematical formulation for the RCC problem. We also discuss alternative models for the relaxed structural balance and the solution of clustering problems associated with these new models. Numerical experiments are carried out with each formulation on a set of benchmark instances available in the literature.  相似文献

ABSTRACT

Older adults who are lesbian, gay, bisexual, or transgender (LGBT) face greater health risks and possibly more costly care because of their reluctance to seek out health and long-term care services because of limited cultural sensitivity of service providers. This is particularly evident in older lesbians who face substantial risk of health problems associated with alcoholism and are less likely to be open with health care providers because of stigma combined with feelings of alienation, stress, and depression. An estimated 4.4 million older adults are predicted to have problems with alcohol by 2020, and the rates of alcohol-related hospitalizations are similar to those for heart attacks, creating exorbitant medical costs. More culturally competent health and long-term care may reduce health care costs by effectively addressing the dynamics of alcoholism, aging, and lesbian culture. Training initiatives such as those developed by the National Resource Center on LGBT Aging have begun to address the need of a more culturally competent aging services network. This article provides exemplars from empirical data on older lesbians with alcoholism to highlight some of the health, economic, and social disparities experienced in the aging LGBT community. Current interventions in the form of cultural competence training for service providers are presented as a potential step toward addressing health disparities among LGBT older adults.  相似文献

This paper questions the usefulness of the rights-based approach to ameliorating the social situation of disabled people in Britain and advances two criticisms. First, that rights and self-defined needs have been under-theorised by disability theorists to the extent that they have insufficiently appreciated the problems that these approaches pose. The paper suggests that rights to appropriate resources to satisfy self-defined needs will generate vast numbers of competing rights claims and that the resulting tendency of rights to conflict has been under-appreciated. Secondly, that there has been little consideration of how these conflicts might be reconciled. The first two sections of the paper look at the concepts of ascribed and self-defined needs, respectively, whilst the final one looks at some of the problems of the rights approach and some of the difficulties of making self-defined need the basis of rights claims.  相似文献

The nominal group technique, a small-group decision-making process which offers open and relatively unrestricted response to problem identification and resolution, was used as a means of identifying the major stressors of elementary school teachers and the concomitant solutions to their problems. Although the specific stressors that have been presented in this paper are not necessarily applicable to other school districts, the nominal group technique itself should be very useful in other school districts as well as in most any work setting.The active involvement of teachers in defining and solving their own problems is a critical aspect of the technique. Teachers, like most employees, will more actively solve problems and accept innovations if they can see some tangible benefit for themselves in doing so.“Intensity of work demands” was the most frequently mentioned job stressor and also received the highest combined ranking for the nine teacher groups. “Student misbehavior and lack of motivation” ranked a close second.  相似文献

Population aging is a phenomenon occurring across the globe including in countries traditionally exhibiting population dividends and “youth bulges.” The Gulf Corporation Council countries are no exception as they currently experience a process of population aging, albeit at a different stage from many developed countries. However, due to historically high fertility rates and fast-paced epidemiological transition, some of these countries will experience population aging at a higher pace than what has been observed in Europe and the United States. This article reviews recent developments in long-term care policies in the Gulf region with a focus on Oman as an example of a high-income Arab country that is experiencing population aging while still being governed by traditional family aged-care norms. Utilizing existing data and published research complemented by policy analysis and field visits, we analyze the process of population aging in Oman and neighboring countries and its policy implications.  相似文献

In recent years, there has been a decline in language skills among the younger population in Sweden. Within several different academic programs, it has been observed that students arriving at the universities do not have the skills needed to make use of what is taught. This raises a number of problems, especially within social work education, because language skills, both written and oral, are important tools for practicing social work. This article presents a way of meeting this challenge and improving the students’ skills by including them in discussions on how to write academically as part of teaching method called language guidance. The students reflect upon their own and other students’ texts, and discuss how they can be improved with regard to spelling, grammar, paragraphing, and clarity. Overall, the resources devoted to improving students’ language skills have resulted in improvements in the essays the student write later on in their education.  相似文献

This paper explores the research evidence from England and France on the mental health of young people aging out of care and into adulthood. It represents the first comparative review based on the evidence from these two countries. Set in the legal, policy and service framework for both countries, it reviews evidence on the mental health of young people in the general population, young people living in care, young people aging out of care, and young adults. It shows: the high levels of psychological adversity of young people entering care; the high rates of mental health problems of young people in care compared with the general population of young people; the increased risk of mental and physical problems at the time of aging out of care, and the general improvement in longer-term outcomes for young adults, although some continue to have serious mental health problems. In conclusion, it suggests that interventions across the life course of young people are needed.  相似文献

The worm Caenorhabditis elegans has become a popular model organism for the study of mechanisms involved in aging. The C. elegans life span is controlled by several pathways that have been extensively characterized at the molecular level. These include pathways that regulate metabolism and development (namely, the insulin/IGF-1 pathway), nutrition, mitochondrial activity, and reproduction. Presentations at a recent C. elegans conference add to the growing body of knowledge about the genetic networks that control the complex process of aging and suggest new avenues for further investigations.  相似文献

Osteoarthritis (OA) is a chronic disease that disables many aging adults. People with OA are often asked to adhere to prescribed exercise regimens that must be undertaken in the presence of pain and other disease-related symptoms. We conducted a review of literature that focused on what is known about exercise adherence and the factors that influence exercise adherence among people with OA. Results revealed multiple determinants of exercise adherence; however, these determinants have not been carefully studied in the context of exercise adherence and OA. Almost all studies of exercise adherence among people with OA are short-term and do not use validated measures of adherence. Moreover, poor adherence is the most compelling explanation for the declining impact of the benefits of exercise over time. We conclude that interventions to enhance self-efficacy, social support, and skills in long-term monitoring of progress are necessary to foster exercise adherence among people with OA.  相似文献

Cells in the body grow and die, cells in lab dishes grow and die, and individual organisms grow and die. The parallels seem maddeningly obvious, but scores of scientists still labor to draw the correct connections, to uncover the mechanisms that underlie aging in cell culture flasks and in whole animals. Do our cells stop growing, quit working, cease dividing, or start dying as we age? Do we die when our cells do, or are we somehow more than the sum of our cells? For decades, scientists have searched for evidence that links changes in cell growth, cell function, cell division, and cell death to the phenomenon we call aging. Although definitive proof eludes them, researchers continue to conduct experiments in tissue culture and in animal models, amassing information that points us toward a greater understanding of what aging is--and is not.  相似文献

Why have social constructionists remained absent from debates over public sociology? I argue that constructionist scholarship would be particularly amenable to Michael Burawoy’s notion of ‘organic’ public sociology, given the ability of constructionist scholars to orient awareness contexts in order to help engender constructionist imaginations. This approach requires that constructionists take on a different view of the role of the analyst. I also discuss some of the problems Canadian academics have had engaging with the media in their efforts to engage in ‘traditional’ public sociology, as well as what a constructionist public sociology may look like practice. I conclude by addressing potential challenges to a constructionist public sociology within Canada, including reference to sociology’s disciplinary coherence and how we can approach—and what we mean by—‘publics’.  相似文献

Neurodegenerative diseases typically affect subpopulations of neurons. Characterizing these vulnerable cells and identifying the factors that make them susceptible to damage while neighboring cells remain resistant are essential to the understanding of molecular pathogenesis that underlies neurodegenerative diseases. Classically, molecular analysis of the central nervous system involves the identification and isolation of an anatomic region of interest; next, the relevant tissue is pulverized, and the resulting homogenate is analyzed. Although this method provides useful data, its effectiveness diminishes when used in areas of high cellular diversity or in instances in which one cell type is lost as a consequence of selective cell death or quiescence. A technique that affords the ability to assess molecular events in a very precise anatomical site would provide a powerful tool for this research discipline. In this review, we discuss the amplification of messenger RNA from single neural cells and the subsequent use of the RNA to probe DNA microarrays in an effort to create cell-specific molecular profiles. Specifically, recent work in single-cell expression profiling in Alzheimer's and Huntington's diseases is discussed. We also review some new work with neural stem cells and their application to restorative neurobiology. Finally, we discuss the use of cell-specific molecular profiles to better understand the basics of neuronal cell biology.  相似文献
