Leptospirosis is a preeminent zoonotic disease concentrated in tropical areas, and prevalent in both industrialized and rural settings. Dose‐response models were generated from 22 data sets reported in 10 different studies. All of the selected studies used rodent subjects, primarily hamsters, with the predominant endpoint as mortality with the challenge strain administered intraperitoneally. Dose‐response models based on a single evaluation postinfection displayed median lethal dose (LD50) estimates that ranged between 1 and 107 leptospirae depending upon the strain's virulence and the period elapsed since the initial exposure inoculation. Twelve of the 22 data sets measured the number of affected subjects daily over an extended period, so dose‐response models with time‐dependent parameters were estimated. Pooling between data sets produced seven common dose‐response models and one time‐dependent model. These pooled common models had data sets with different test subject hosts, and between disparate leptospiral strains tested on identical hosts. Comparative modeling was done with parallel tests to test the effects of a single different variable of either strain or test host and quantify the difference by calculating a dose multiplication factor. Statistical pooling implies that the mechanistic processes of leptospirosis can be represented by the same dose‐response model for different experimental infection tests even though they may involve different host species, routes, and leptospiral strains, although the cause of this pathophysiological phenomenon has not yet been identified.  相似文献

Interspecies Extrapolation: A Reexamination of Acute Toxicity Data   总被引:2,自引:0,他引:2  
We reanalyze the acute toxicity data on cancer chemotherapeutic agents compiled by Freireich et al.(1) and Schein et al.(2) to derive coefficients of the allometric equation for scaling toxic doses across species (toxic dose = a.[body weight]b). In doing so, we extend the analysis of Travis and White (Risk Analysis, 1988, 8, 119-125) by addressing uncertainties inherent in the analysis and by including the hamster data, previously not used. Through Monte Carlo sampling, we specifically account for measurement errors when deriving confidence intervals and testing hypotheses. Two hypotheses are considered: first, that the allometric scaling power (b) varies for chemicals of the type studied; second, that the same scaling power, or "scaling law," holds for all chemicals in the data set. Following the first hypothesis, in 95% of the cases the allometric power of body weight falls in the range from 0.42-0.97, with a population mean of 0.74. Assuming the second hypothesis to be true-that the same scaling law is followed for all chemicals-the maximum likelihood estimate of the scaling power is 0.74; confidence bounds on the mean depend on the size of measurement error assumed. Under a "best case" analysis, 95% confidence bounds on the mean are 0.71 and 0.77, similar to the results reported by Travis and White. For alternative assumptions regarding measurement error, the confidence intervals are larger and include 0.67, but not 1.00. Although a scaling power of about 0.75 provides the best fit to the data as a whole, a scaling power of 0.67, corresponding to scaling per unit surface area, is not rejected when the nonhomogeneity of variances is taken into account. Hence, both surface area and 0.75 power scaling are consistent with the Freireich et al. and Schein et al. data sets. To illustrate the potential impact of overestimating the scaling power, we compare reported human MTDs to values extrapolated from mouse LD10s.  相似文献

Differences between American and Japanese workers regarding perceptions of effective leadership were investigated. 128 American workers and 203 Japanese workers were asked to describe the characteristics of their ideal leader. Results indicated that Americans perceive personality characteristics as more important for leadership, whereas Japanese perceive skills and behaviors as more important for leadership. Suggestions are made for training leaders from the U.S. and Japan for cross-cultural situations and for future cross-cultural leadership research.  相似文献

Complex systems often involve a huge number of variables whose interdependence must be taken into account. This makes the adoption of an integrated approach essential for the proper management of a wide range of aspects related to human life, from social matters to the environment. In particular, this paper takes into consideration three different experiences in which the use of an integrated approach represented the most effective response for managing the evolving realities we took into consideration. We focused on the issue of land management, with particular attention to the problem of monoculture in Brazil. Then we analyzed the attempted cooperation between the States of the Aral Sea area for efficient water management of the basin and, finally, we dealt with the matter of energy efficiency and how the production of home integrated systems could lead to a better rationalization of energy consumption in the near future.   相似文献

ABSTRACT

Fisher's linear discriminant analysis (FLDA) is known as a method to find a discriminative feature space for multi-class classification. As a theory of extending FLDA to an ultimate nonlinear form, optimal nonlinear discriminant analysis (ONDA) has been proposed. ONDA indicates that the best theoretical nonlinear map for maximizing the Fisher's discriminant criterion is formulated by using the Bayesian a posterior probabilities. In addition, the theory proves that FLDA is equivalent to ONDA when the Bayesian a posterior probabilities are approximated by linear regression (LR). Due to some limitations of the linear model, there is room to modify FLDA by using stronger approximation/estimation methods. For the purpose of probability estimation, multi-nominal logistic regression (MLR) is more suitable than LR. Along this line, in this paper, we develop a nonlinear discriminant analysis (NDA) in which the posterior probabilities in ONDA are estimated by MLR. In addition, in this paper, we develop a way to introduce sparseness into discriminant analysis. By applying L1 or L2 regularization to LR or MLR, we can incorporate sparseness in FLDA and our NDA to increase generalization performance. The performance of these methods is evaluated by benchmark experiments using last_exam17 standard datasets and a face classification experiment.  相似文献

Abstract  This study examines the relationship between workers' lifestyle, organizational commitment, and satisfaction. First, this study describes the lifestyle, i.e. cluster of value orientations, of male workers in Tokyo. The sample of this study is 2,003 regular male private firm employees and public (government) employees aged 20-54 living in the Tokyo metropolitan areas. We measure attitudes of workers toward work and the place of work. The 33 measures of attitudes are analyzed by means of cluster analysis to construct typologies of lifestyle of workers. We obtained three clusters: cluster 1 (career-oriented generalists, i.e., company man), cluster 2 (stability-oriented family man), and cluster 3 (work-oriented specialist). Second, we found demographic differences among the three clusters. Workers in cluster 1 tend to be older. to be more university educated, to obtain higher income, and to have higher positions at their current job. Workers in cluster 2 are more likely to be young, to graduate from senior high school, and to be in the middle-income group. Workers in cluster 3 tend to graduate from special training school or university and to be in lower-income group. Finally, we found differences in organizational commitment and job satisfaction among the three clusters. Workers in cluster 2 are more likely than are those in the other two clusters to be dissatisfied with their current job. to have a lower level of identification with their current firm, and consequently to have a greater propensity to leave their present company.  相似文献
