Reduced-rank regression models proposed by Anderson [1951. Estimating linear restrictions on regression coefficients for multivariate normal distributions. Ann. Math. Statist. 22, 327–351] have been used in various applications in social and natural sciences. In this paper we combine the features of these models with another popular, seemingly unrelated regression model proposed by Zellner [1962. In an observational study in which each treated subject is matched to several untreated controls by using observed pretreatment covariates, a sensitivity analysis asks how hidden biases due to unobserved covariates might alter the conclusions. The bounds required for a sensitivity analysis are the solution to an optimization problem. In general, this optimization problem is not separable, in the sense that one cannot find the needed optimum by performing a separate optimization in each matched set and combining the results. We show, however, that this optimization problem is asymptotically separable, so that when there are many matched sets a separate optimization may be performed in each matched set and the results combined to yield the correct optimum with negligible error. This is true when the Wilcoxon rank sum test or the Hodges-Lehmann aligned rank test is applied in matching with multiple controls. Numerical calculations show that the asymptotic approximation performs well with as few as 10 matched sets. In the case of the rank sum test, a table is given containing the separable solution. With this table, only simple arithmetic is required to conduct the sensitivity analysis. The method also supplies estimates, such as the Hodges-Lehmann estimate, and confidence intervals associated with rank tests. We consider the estimation of Poisson regression models in which structural variation in a subset of the parameters is permitted. It is noted that coventional estimation algorithms are likely to impose restrictions on the number of explanatory variables and the number of structural regimes. We propose an alternative algorithm that implements partitioned matrix inversion and thereby avoids restictions on the size of the model. Selecting a small subset out of the thousands of genes in microarray data is important for accurate classification of phenotypes. In this paper, we propose a flexible rank-based nonparametric procedure for gene selection from microarray data. In the method we propose a statistic for testing whether area under receiver operating characteristic curve (AUC) for each gene is equal to 0.5 allowing different variance for each gene. The contribution to this “single gene” statistic is the studentization of the empirical AUC, which takes into account the variances associated with each gene in the experiment. Delong et al. proposed a nonparametric procedure for calculating a consistent variance estimator of the AUC. We use their variance estimation technique to get a test statistic, and we focus on the primary step in the gene selection process, namely, the ranking of genes with respect to a statistical measure of differential expression. Two real datasets are analyzed to illustrate the methods and a simulation study is carried out to assess the relative performance of different statistical gene ranking measures. The work includes how to use the variance information to produce a list of significant targets and assess differential gene expressions under two conditions. The proposed method does not involve complicated formulas and does not require advanced programming skills. We conclude that the proposed methods offer useful analytical tools for identifying differentially expressed genes for further biological and clinical analysis. 相似文献

Sample size re-estimation without unblinding for normally distributed outcomes with unknown variance
Monitoring clinical trials in nonfatal diseases where ethical considerations do not dictate early termination upon demonstration of efficacy often requires examining the interim findings to assure that the protocol-specified sample size will provide sufficient power against the null hypothesis when the alternative hypothesis is true. The sample size may be increased, if necessary to assure adequate power. This paper presents a new method for carrying out such interim power evaluations for observations from normal distributions without unblinding the treatment assignments or discernably affecting the Type 1 error rate.
