In a response-adaptive design, we review and update the trial on the basis of outcomes in order to achive a specific goal. In clinical trials our goal is to allocate a larger number of patients to the better treatment. In the present paper, we use a response adaptive design in a two-treatment two-period crossover trial where the treatment responses are continuous. We provide probability measures to choose between the possible treatment combinations AA, AB, BA, or BB. The goal is to use the better treatment combination a larger number of times. We calculate the allocation proportions to the possible treatment combinations and their standard errors. We also derive some asymptotic results and provide solutions on related inferential problems. The proposed procedure is compared with a possible competitor. Finally, we use a data set to illustrate the applicability of our proposed design.  相似文献

The randomized response (RR) technique with two decks of cards proposed by Odumade and Singh (2009) can always be made more efficient than the RR techniques proposed by Warner (1965), Mangat and Singh (1990), and Mangat (1994) by adjusting the proportion of cards in the decks. Arnab et al. (2012) generalized Odumade and Singh strategy (2009) for complex survey designs and wider class of estimators. In this paper improvement of Arnab et al. Results from classical linear regression regarding the effects of covariate adjustment, with respect to the issues of confounding, the precision with which an exposure effect can be estimated, and the efficiency of hypothesis tests for no treatment effect in randomized experiments, are often assumed to apply more generally to other types of regression models. In this paper results pertaining to several generalized linear models involving a dichotomous response variable are given, demonstrating that with respect to the issues of confounding and precision, for models having a linear or log link function the results of classical linear regression do generally apply, whereas for other models, including those having a logit, probit, log-log, complementary log-log, or generalized logistic link function, the results of classical linear regression do not always apply. It is also shown, however, that for any link function, covariate adjustment results in improved efficiency of hypothesis tests for no treatment effect in randomized experiments, and hence that the classical linear regression results regarding efficiency do apply for all models having a dichotomous response variable.  相似文献

We propose an efficient group sequential monitoring rule for clinical trials. At each interim analysis both efficacy and futility are evaluated through a specified loss structure together with the predicted power. The proposed design is robust to a wide range of priors, and achieves the specified power with a saving of sample size compared to existing adaptive designs. A method is also proposed to obtain a reduced-bias estimator of treatment difference for the proposed design. The new approaches hold great potential for efficiently selecting a more effective treatment in comparative trials. Operating characteristics are evaluated and compared with other group sequential designs in empirical studies. An example is provided to illustrate the application of the method.  相似文献

Brownian motion has been used to derive stopping boundaries for group sequential trials, however, when we observe dependent increment in the data, fractional Brownian motion is an alternative to be considered to model such data. In this article we compared expected sample sizes and stopping times for different stopping boundaries based on the power family alpha spending function under various values of Hurst coefficient. Results showed that the expected sample sizes and stopping times will decrease and power increases when the Hurst coefficient increases. With same Hurst coefficient, the closer the boundaries are to that of O'Brien-Fleming, the higher the expected sample sizes and stopping times are; however, power has a decreasing trend for values start from H = 0.6 (early analysis), 0.7 (equal space), 0.8 (late analysis). We also illustrate study design changes using results from the BHAT study.  相似文献

This study investigates the small sample powers of several tests designed against ordered location alternatives in randomized block experiments. The results are intended to aid the researcher in the selection process. Toward this end the small sample powers of three classes of rank tests — tests based on ‘within-blocks’ rankings (W-tests), ‘among-b locks’ rankings (A-tests), and ‘ranking after alignment’ within blocks (RAA-tests)— are compared and contrasted with the asymptotic properties given by Pirie (1974) as well as with the empirical powers of competing parametric procedures.  相似文献

Formal inference in randomized clinical trials is based on controlling the type I error rate associated with a single pre‐specified statistic. The deficiency of using just one method of analysis is that it depends on assumptions that may not be met. For robust inference, we propose pre‐specifying multiple test statistics and relying on the minimum p‐value for testing the null hypothesis of no treatment effect. The null hypothesis associated with the various test statistics is that the treatment groups are indistinguishable. The critical value for hypothesis testing comes from permutation distributions. Rejection of the null hypothesis when the smallest p‐value is less than the critical value controls the type I error rate at its designated value. Even if one of the candidate test statistics has low power, the adverse effect on the power of the minimum p‐value statistic is not much. Its use is illustrated with examples. We conclude that it is better to rely on the minimum p‐value rather than a single statistic particularly when that single statistic is the logrank test, because of the cost and complexity of many survival trials. 网络道德审判是对传统道德审判的继承与发展,存在着显著的伦理困境:它理应通过激发正向羞耻感来促进对行为者的伦理教化,但在诸多情况中却因引发负向羞耻感而产生对行为者的伦理排斥。网络道德审判作为一种典型的道德评价形态,可以利用情感主义伦理学中的旁观者视角来加以考察。据此可发现,网络旁观者的同情、想象、事实判断能力存在着诸多困境,同时其群体性特征又进一步加剧了潜在的风险。实现公正的网络道德审判,需要汲取和借鉴“公正的旁观者”的多重意蕴来构建可能的伦理进路。  相似文献

Abstract

Conclusions of sample surveys dealing with matters of privacy are often disputed. Many people either refuse to participate or provide false answers. Indirect questioning techniques are designed so that reliable estimates can be produced while the privacy of the participants is protected. One such method, the Item Count Technique is gaining increasing popularity. However, the original version of the method does not fully protect the privacy of the participants. In this paper we propose a new version of the technique which better protects the privacy of the respondents and does not have the so-called floor and ceiling effects.  相似文献

For a controlled branching process (CBP) with offspring distribution belonging to the power series family, the asymptotic normality of the posterior distribution of the basic parameter and the offspring mean is proved. As practical applications, we calculate asymptotic high probability density credibility sets for the offspring mean and we provide a rule to make inference about the value of this parameter. Moreover, the asymptotic posterior normality of the respective parameters of two classical branching models, namely the standard Galton–Watson process and the Galton–Watson process with immigration, is derived as particular cases of the CBP.  相似文献
