With the increasing use of nanomaterials incorporated into consumer products, there is a need for developing approaches to establish “quantitative structure‐activity relationships” (QSARs). These relationships could be used to predict various biological responses after exposure to nanomaterials for the purposes of risk analysis. This risk analysis is applicable to manufacturers of nanomaterials in an effort to determine potential hazards. Because metal oxide materials are some of the most widely applicable and studied nanoparticle types for incorporation into cosmetics, food packaging, and paints and coatings, we focused on comparing different approaches for establishing QSARs for this class of materials. Metal oxide nanoparticles are believed, by some, to cause alterations in cellular function due to their size and/or surface area. Others have said that these nanomaterials, because of the oxidized state of the metal, do not induce stress in biological tests systems. This controversy highlights the need to systematically develop structure‐activity relationships (i.e., the relationship between physicochemical features to the cellular responses) and tools for predicting potential biological effects after a metal oxide nanomaterial exposure. Here, we attempt to identify a set of properties of two specific metal oxide nanomaterials—TiO2 and ZnO—that could be used to characterize and predict the induced cellular membrane damage of immortalized human lung epithelial cells. We adopt a mathematical modeling approach that uses the engineered nanomaterial size characterized as a dry nanopowder and the nanomaterial behavior in ultrapure water, phosphate buffer, and cell culture media to predict nanomaterial‐induced cellular membrane damage (via lactate dehydrogenase release). Results of these studies provide insights on how engineered nanomaterial features influence cellular responses and thereby outline possible approaches for developing and applying predictive computational models for biological responses caused by exposure to nanomaterials. 相似文献

This article introduces a regularized logistic discrimination method that is especially suited for discretized stochastic processes (such as periodograms, spectrograms, EEG curves, etc.). The proposed method penalizes the total variation of the discriminant directions, giving smaller misclassification errors than alternative methods, and smoother and more easily interpretable discriminant directions. This paper considers the general problem of Feasible Generalized Least Squares Instrumental Variables (FGLS IV) estimation using optimal instruments. First we summarize the sufficient conditions for the FGLS IV estimator to be asymptotically equivalent to an optimal GLS IV estimator. Then we specialize to stationary dynamic systems with stationary VAR errors, and use the sufficient conditions to derive new moment conditions for these models. These moment conditions produce useful IVs from the lagged endogenous variables, despite the correlation between errors and endogenous variables. This use of the information contained in the lagged endogenous variables expands the class of IV estimators under consideration and thereby potentially improves both asymptotic and small-sample efficiency of the optimal IV estimator in the class. Some Monte Carlo experiments compare the new methods with those of Hatanaka (1976). For the DGP used in the Monte Carlo experiments, asymptotic efficiency is strictly improved by the new IVs, and experimental small-sample efficiency is improved as well. 相似文献

The area under the ROC curve (AUC) can be interpreted as the probability that the classification scores of a diseased subject is larger than that of a non-diseased subject for a randomly sampled pair of subjects. From the perspective of classification, we want to find a way to separate two groups as distinctly as possible via AUC. When the difference of the scores of a marker is small, its impact on classification is less important. Thus, a new diagnostic/classification measure based on a modified area under the ROC curve (mAUC) is proposed, which is defined as a weighted sum of two AUCs, where the AUC with the smaller difference is assigned a lower weight, and vice versa. Using mAUC is robust in the sense that mAUC gets larger as AUC gets larger as long as they are not equal. Moreover, in many diagnostic situations, only a specific range of specificity is of interest. Under normal distributions, we show that if the AUCs of two markers are within similar ranges, the larger mAUC implies the larger partial AUC for a given specificity. This property of mAUC will help to identify the marker with the higher partial AUC, even when the AUCs are similar. Two nonparametric estimates of an mAUC and their variances are given. We also suggest the use of mAUC as the objective function for classification, and the use of the gradient Lasso algorithm for classifier construction and marker selection. Application to simulation datasets and real microarray gene expression datasets show that our method finds a linear classifier with a higher ROC curve than some other existing linear classifiers, especially in the range of low false positive rates. 相似文献

To assess independence in two-way contingency tables, the Pearson chi-square test or Fisher’s exact test are typically used. These tests assume that each subject contributes at most one count to only one table cell (e.g., sex versus blood type). In other situations, each subject may have more than one count contributing to the table and these counts may occur in different cells of the table. One may wish to test independence, adjusting for the within-subject correlation. We provide a simple nonparametric bootstrap approach and assess its performance through simulation studies.
