One of the most dynamic and fruitful areas of current health‐related research concerns the various roles of the human microbiome in disease. Evidence is accumulating that interactions between substances in the environment and the microbiome can affect risks of disease, in both beneficial and adverse ways. Although most of the research has concerned the roles of diet and certain pharmaceutical agents, there is increasing interest in the possible roles of environmental chemicals. Chemical risk assessment has, to date, not included consideration of the influence of the microbiome. We suggest that failure to consider the possible roles of the microbiome could lead to significant error in risk assessment results. Our purpose in this commentary is to summarize some of the evidence supporting our hypothesis and to urge the risk assessment community to begin considering and influencing how results from microbiome‐related research could be incorporated into chemical risk assessments. An additional emphasis in our commentary concerns the distinct possibility that research on chemical–microbiome interactions will also reduce some of the significant uncertainties that accompany current risk assessments. Of particular interest is evidence suggesting that the microbiome has an influence on variability in disease risk across populations and (of particular interest to chemical risk) in animal and human responses to chemical exposure. The possible explanatory power of the microbiome regarding sources of variability could reduce what might be the most significant source of uncertainty in chemical risk assessment.  相似文献

In many toxicological assays, interactions between primary and secondary effects may cause a downturn in mean responses at high doses. In this situation, the typical monotonicity assumption is invalid and may be quite misleading. Prior literature addresses the analysis of response functions with a downturn, but so far as we know, this paper initiates the study of experimental design for this situation. A growth model is combined with a death model to allow for the downturn in mean doses. Several different objective functions are studied. When the number of treatments equals the number of parameters, Fisher information is found to be independent of the model of the treatment means and on the magnitudes of the treatments. In general, A- and DA-optimal weights for estimating adjacent mean differences are found analytically for a simple model and numerically for a biologically motivated model. Results on c-optimality are also obtained for estimating the peak dose and the EC50 (the treatment with response half way between the control and the peak response on the increasing portion of the response function). Finally, when interest lies only in the increasing portion of the response function, we propose composite D-optimal designs.  相似文献

Estimating Consumer Exposure to PFOS and PFOA   总被引:4,自引:0,他引:4  
Perfluorinated compounds have been used for more than 50 years as process aids, surfactants, and for surface protection. This study is a comprehensive assessment of consumer exposure to perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA) from a variety of environmental and product-related sources. To identify relevant pathways leading to consumer exposure to PFOS and PFOA a scenario-based approach has been applied. Scenarios represent realistic situations where age- and gender-specific exposure occurs in the everyday life of consumers. We find that North American and European consumers are likely to experience ubiquitous and long-term uptake doses of PFOS and PFOA in the range of 3 to 220 ng per kg body weight per day (ng/kgbw/day) and 1 to 130 ng/kgbw/day, respectively. The greatest portion of the chronic exposure to PFOS and PFOA is likely to result from the intake of contaminated foods, including drinking water. Consumer products cause a minor portion of the consumer exposure to PFOS and PFOA. Of these, it is mainly impregnation sprays, treated carpets in homes, and coated food contact materials that may lead to consumer exposure to PFOS and PFOA. Children tend to experience higher total uptake doses (on a body weight basis) than teenagers and adults because of higher relative uptake via food consumption and hand-to-mouth transfer of chemical from treated carpets and ingestion of dust. The uptake estimates based on scenarios are within the range of values derived from blood serum data by applying a one-compartment pharmacokinetic model.  相似文献

This study proposed and tested a model of factors influencing the effectiveness of Local Emergency Planning Committees (LEPCs). Data were collected from the chairs of Michigan LEPCs, who were asked to provide information about their LEPCs’staffing, activities, resources, and team climate, as well as information about community hazards and relationships with other organizations in their communities. The model was partially supported. As hypothesized, team climate substantially mediated the relationships between an LEPCs effectiveness and many of the attributes of its internal structure and external environment. In addition, three (emergency planning resources, community support and LEPC staffing and structure) out of four categories of variables hypothesized to influence team climate were, in fact directly related to it. There were two major inconsistencies with the model. First, two types of emergency planning resources were found to have a direct influence on LEPC effectiveness rather than an indirect effect mediated by team climate. In addition, hazard vulnerability was hypothesized to have a direct effect on team climate, but the data showed this relationship was mediated by community support and by LEPC staffing and structure. A revised model was proposed that suggests ways in which members can change the structure of an LEPC and strategies to enhance its effectiveness.  相似文献

Risk perception may be influenced by a number of factors, such as unfamiliarity, lack of control, perceived consequences, and hazards being seen as catastrophic and having risk for future generations. Risk perception researchers have typically used such investigator-selected characteristics to assess hazards. In the first study reported here, the repertory grid method was used to elicit the terminology that subjects ( n = 30) use to distinguish between 30 different chemicals. The data were submitted to generalized Procrustes analysis. The first principal axis of the resulting consensus plot separated the chemicals ranging from "poisonous or toxic,""harmful or dangerous," and "sounds negative" at one end, to "positive effect on health,""often present in food nowadays," and "sounds positive" at the other end. The second principal axis ranged from "familiar with or knowledge of" and "chemical" to "natural." A second study ( n = 226) was carried out to look at the general validity of the results of the repertory grid interviews using a fixed questionnaire. The data were submitted to principal components analysis and internal preference mapping. The first principal component ranged from "safe" and "healthy" at one end, to "poisonous" and "harmful" at the other end. The chemicals also separated in terms of "familiar,""chemical," and "natural." All three methods of data collection and analysis yield essentially similar results.  相似文献

面对危化品危害,政府及相关单位面临着日益严峻的既要保证公众知情,又要避免信息过度传播的难题,"信任"是导致这种沟通困境的重要因素。为解决危害信息有效沟通问题,从重构公众与政府之间信任关系的角度入手,以公众利益相关者感知衡量信任,探究公众社区参与意愿对利益相关者感知的影响关系,并将社区参与分为代表单向沟通的仪式性参与和代表双向沟通的实质性参与。研究结果表明:公众社区参与意愿有利于提升公众危化品危害利益相关者感知,特别是仪式性参与意愿的直接作用效果更为显著;实质性参与意愿和危害利益相关者感知的关系中,预决策过程起到完全中介的作用;危化品危害知识在社区参与意愿和利益相关者感知的影响关系中起到显著的正向调节作用,特别是在预决策过程与利益相关者感知的影响路径中。研究结论为构建公众和政府之间的信任关系,进而为缓解危化品危害信息沟通难题提供理论支撑和实践价值。  相似文献

This article describes the development and application of a generic approach to the comparative assessment of risks related to the production of organic chemicals by petrochemical processes versus white biotechnology. White biotechnology, also referred to as industrial biotechnology, typically uses bio-based feedstocks instead of the fossil raw materials used in the petrochemical sector. The purpose of this study was to investigate whether the production of chemicals by means of white biotechnology has lower conventional risks than their production by petrochemical processes. Conventional risks are the risks of well-established processes, and not those related to genetically modified microorganisms and plants. Our approach combines classical risk assessment methods (largely based on toxicology), as developed by the life cycle assessment (LCA) community, with statistics on technological disasters, accidents, and work-related illnesses. Moreover, it covers the total process chain for both petrochemical and bio-based products from cradle to grave. The approach was applied to five products: the plastics polytrimethylene terephthalate (PTT), polyhydroxyalkanoates (PHA), polyethylene terephthalate (PET), polyethylene (PE), and ethanol. Our results show that the conventional risks related to the white biotechnology products studied are lower than those of the petrochemical products. However, considering the uncertainties with respect to the ranges of input data, the (incomplete) coverage of emissions by the environmental priority strategies (EPS) 2000 method, and the uncertainties of the assumptions made in this study (i.e., large to very large), the differences in results between bio-based and petrochemical products fall into the uncertainty range. Because of this, future research is necessary to decrease the uncertainties before we can conclude that the conventional risks of biotechnologically produced chemicals are lower than those of fossil-fuel-derived chemicals.  相似文献

Chemicals indisputably contribute greatly to the well-being of modern societies. Apart from such benefits, however, chemicals often pose serious threats to human health and the environment when improperly handled. Therefore, the European Commission has proposed a regulatory framework for the Registration, Evaluation and Authorization of Chemicals (REACH) that requires companies using chemicals to gather pertinent information on the properties of these substances. In this article, we argue that the crucial aspect of this information management may be the honesty and accuracy of the transfer of relevant knowledge from the producer of a chemical to its user. This may be particularly true if the application of potentially hazardous chemicals is not part of the user's core competency. Against this background, we maintain that the traditional sales concept provides no incentives for transferring this knowledge. The reason is that increased user knowledge of a chemical's properties may raise the efficiency of its application. That is, excessive and unnecessary usage will be eliminated. This, in turn, would lower the amount of chemicals sold and in competitive markets directly decrease profits of the producer. Through the introduction of chemical leasing business models, we attempt to present a strategy to overcome the incentive structure of classical sales models, which is counterproductive for the transfer of knowledge. By introducing two models (a Model A that differs least and a Model B that differs most from traditional sales concepts), we demonstrate that chemical leasing business models are capable of accomplishing the goal of Registration, Evaluation and Authorization of Chemicals: to effectively manage the risk of chemicals by reducing the total quantity of chemicals used, either by a transfer of applicable knowledge from the lessor to the lessee (Model A) or by efficient application of the chemical by the lessor him/herself (Model B).  相似文献
