黏附细胞的超微结构包括细胞骨架(cytoskeleton)、细胞黏附点(focal adhesion,FA)及细胞间连接(gap junction,GJ).保持细胞超微结构的完整,对复温后细胞的存活率及细胞功能是至关重要的.实验发现,冻结到一196℃后,部分F肌动蛋白被切断、卷曲,且扩展面积与细胞成活率随温度降低呈现相似的下降趋势;冻结后蛋白质vinculin(FA)和Cx-43(GJ)的数量分别下降到对照组的22%及10%左右.综述了冻结过程对细胞超微结构影响的研究现状,分析了目前的研究所存在的问题与不足,提出了实时观察活细胞超微结构的方法,并给出了初步的结果. 相似文献

Clinical implications and ethical dilemmas of the use of confidential case material in clinical writing are examined, including
a review of the discourse among professionals who publish clinical work. This literature is applied to a clinical illustration

Siblings who have suffered severe trauma and disrupted attachment relate to one another in ways that interfere with individual development, the chance to benefit from connections with adults, the establishment of healthy sibling relationships, and the working through of traumatic experiences. The authors use case examples to illustrate four distinct sibling patterns: absent, adult lockout, half and half, and trauma shield. The authors highlight the importance of recognizing these types and then varying the therapist's treatment stance to pursue five goals. These include suspending problematic sibling relationships to create room for bonding with caring adults, transferring attachment behaviors and impulses to a receptive adult, resuming healthy individual development, forming more adaptive sibling connections, and expressing shared traumatic content. 相似文献

Attachment‐based research has flourished over the past few years and it now includes an impressive array of studies across the lifespan. This article considers four different measures used in attachment‐based research, although more discussion is devoted to interview and projective measures because they specifically embrace a psychodynamic approach in their design and analytic focus. Although attachment security has been associated with children's rule‐compatible conduct, the mechanism through which attachment influences early regard for rules is not well established. We hypothesized that effortful control would mediate the link between security and indicators of children's emerging regard for rules (discomfort following rule violations, internalization of parents’ and experimenter's rules, few externalizing behaviors). In a longitudinal study, the Attachment Q‐Set was completed by parents, effortful control was observed, and Regard for Rules was observed and rated by parents. The proposed model fit the data well: Children's security to mothers predicted their effortful control, which in turn had a direct link to a greater Regard for Rules. Children's security with fathers did not predict effortful control. The mother‐child relationship appears particularly important for positive developmental cascades of self‐regulation and socialization. 相似文献

Social capital has been considered a cause and consequence of various uses of new information and communication technologies (ICTs). However, there is a growing divergence between how social capital is commonly measured in the study of ICTs and how it is measured in other fields. This departure raises questions about the validity of some of the most widely cited studies of social capital and ICTs. We compare the Internet Social Capital Scales (ISCS) developed by Williams [2006. On and off the ’net: scales for social capital in an online era. Journal of Computer-Mediated Communication, 11(2), 593–628. doi: 10.1111/j.1083-6101.2006.00029.x] – a series of psychometric scales commonly used to measure ‘social capital’ – to established, structural measures of social capital: name, position, and resource generators. Based on a survey of 880 undergraduate students (the population to which the ISCS has been most frequently administered), we find that, unlike structural measures, the ISCS does not distinguish between the distinct constructs of bonding and bridging social capital. The ISCS does not have convergent validity with structural measures of bonding or bridging social capital; it does not measure the same concept as structural measures. The ISCS conflates social capital with the related constructs of social support and attachment. The ISCS does not measure perceived or actual social capital. These findings raise concerns about the interpretations of existing studies of ‘social capital’ and ICTs that are based on the ISCS. Given the absence of measurement validity, we urge those studying social capital to abandon the ISCS in favor of alternative approaches. 相似文献

The author integrated attachment theory with social cognitive career theory (SCCT) to examine how attachment styles influence the career search self‐efficacy activities of interviewing, networking, job searching, and personal exploration. Using an undergraduate sample (N = 275, 81% women), the author tested a structural model. Results indicated that anxious attachment style was negatively related to interviewing self‐efficacy, whereas avoidant attachment style was negatively related to networking, job searching, and personal exploration self‐efficacy. Theoretical implications based on these findings provide support for an integrative model of attachment theory with SCCT. Career counselors are encouraged to consider clients’ attachment styles when working with individuals who are searching for employment. To help address the limitations of the study, future researchers could test the applicability of the current model with a more diverse sample. 相似文献

Child‐caregiver Attachment Representations in a Non‐Western Context: The Feasibility and Cultural Equivalence of Story Stems in Urban Ghana 下载免费PDF全文
Story stem measures are an increasingly popular method for assessing the attachment representations of young children, but little is known of their cross‐cultural applicability. This study aimed to characterise the attachment representations in 73 five‐ to eight‐year‐old children in urban Ghana, West Africa, using the Manchester Child Attachment Story Task (MCAST) to test its feasibility, psychometric characteristics and concurrent associations with caregiver‐ and teacher‐rated child behaviour, and to conduct a qualitative thematic analysis of methodological observations. Among the classifiable cases (92 percent), all attachment classifications were observed, yielding a higher rate of secure attachment than in European samples. Inter‐rater reliability, internal consistency, and internal structure were reasonable and largely similar to European studies, although one structural difference was the separation of ‘child assuagement of distress’ from other secure‐related items. MCAST narratives were associated with teacher‐ and caregiver‐rated hyperactivity, but internal consistency was low in most Strengths and Difficulties Questionnaire scales. Possible culturally‐sensitive explanations for our psychometric and qualitative findings are discussed. Overall, story stems are a promising tool for accessing attachment representations in non‐Western samples, although modifications are likely to improve cross‐cultural equivalence when applied to non‐Western cultures. Further investigation is needed to link MCAST outcomes to parenting and socio‐emotional development. 相似文献
