艾灸对长期大负荷训练大鼠血睾酮、LH水平和StARmRNA表达的影响

摘    要:目的:探讨艾灸三阴穴交对长期大负荷训练大鼠睾酮(T)的合成作用、血清黄体生成素(LH)水平以及类固醇激素合成急性调节蛋白(StARmRNA)表达的干预效果.方法:雄性SD大鼠30只随机分成三组:对照组、运动组、实验组.除对照组外其它两组每周按照训练方案进行跑台训练建立长期大负荷训练模型,实验组于每天运动2小时后艾灸双侧三阴交穴,对照组和运动组不做艾灸处理,并且每周末记录备组大鼠体重.5周后麻醉组织取材,取血测定红蛋白(HGB),利用放免法检测各组大鼠血清T、LH,RT—PCR检测睾丸组织中stARtuRNA表达水平.结果:(1)5周后,与对照组比较,运动组HGB、血清T水平显著降低(P〈0.05),LH水平无显著差异(P〉0.05),体重随着周次增长变小(P〈0.01),StARmRNA表达量低(P〈0.01).(2)与运动组比较,实验组HGB、血清T水平显著升高(P〈0.05),LH水平无显著差异(P〉0.05),体重随着周次增长变大(P〈0.05),StARmR-NA表达增高(P〈0.05).结论:长时间大负荷的运动训练会使血睾酮水平下降、降低stAR的表达;艾灸三阴交穴可改善长期大负荷训练导致的低血睾酮,可以促进睾丸组织StARmRNA表达,有利于提高运动能力.

Abstract:Objective:To investigate the effect of Moxibusting Sanyinjiao on serum testoserone , LH content and StAR mRNA expression in rats during the long--term and high loading training. Methods: Thirty male rats were divided into three group randomly :control group , exercise group and experimental group . All the groups except control group followed a protocol of treadmill training,experimental group were Moxibusting Sanyinjiao with the corresponding content after two hours, at the same time. control group and exercise group didn't take Moxibustion. weight of rats were recorded on the weeken during the training. After 5--week training, all the rats were taken anesthesia for tissue sampling. Contents of HGB, T, and LH were determined, StAR mRNA expressions of the rats'testicular tissue were detected by PT--PCR. Results:Compared with the control group After five weeks, HGB and serum T Contents of ex- ercise group significantly decreased( P 〈0.05), and StAR mRNA expressions of the rats'testicular tissue significantly decreased,change of the mean body weights became less with the week number increasing( P 〈0.05) ; Compared with the exercise group, HGB and serum T Contents of experimental group signifi- cantly increased( P 〈 0.05), StAR mRNA expressions significantly increased (P〈0.05), change of the mean body weights obviously became greater with the week number increasing( P 〈0.05). Conclusion: It shows that moxibustion can heighten elevate StAR mRNA expressions of the rats'testicular tissue, which improves symptoms of the low serum testoserone.
