补充支链氨基酸对失眠老年大鼠行为和糖代谢的影响

摘    要:目的探讨补充支链氨基酸(branched-chain amino acids,BCAA)对失眠老年大鼠行为和糖代谢的影响。方法采用小平台水环境的睡眠剥夺方法建立动物失眠模型,30只雄性老年大鼠随机分为对照组(C组)、6h组(每天6小时睡眠组)、6h B组(每天睡眠6小时,进食添加BCAA的饲料),4h组(每天睡眠4小时),4h B组(每天睡眠4小时,进食添加BCAA的饲料)。结果实验组大鼠的修饰行为次数、中央格停留时间、粪便颗粒数均高于C组,差异具有显著性(P〈0.05)。且修饰行为次数、中央格停留时间4h B组〈4h组(P〈0.05);旷场实验得分6h组〉C组〉4h组,差异具有显著性(P〈0.05)。但旷场实验得分、粪便颗粒数6h组与6h B组,4h组与4h B组比较无差异(P〉0.05)。与C组比较,实验组的FPG水平显著升高(P〈0.05);但4h B组〈4h组(P〈0.05)。与C组比较4h组和4h B组的血清INS水平显著升高(P〈0.05),且4h B组〈4h组(P〈0.05)。与C组比较,实验组的ISI显著下降;且4h B组〉4h组(P〈0.05)。结论补充BCAA可以改善失眠老年大鼠的行为和糖代谢。

Effects of branched-chain amino acids supplementation on behaviour and glucose metabolism in aged rats subjected to insomnia

Abstract:Objectives To explore the effects of branched-chain amino acids( BCAA) supplementation on behaviour and glucose metabolism in aged rats subjected to isomnia.Methods Experimental model of isomnia was established by use of sleep deprivation method(flower pot),thirty aged male rats were divided into five groups randomly:C(control)group, 6h(6 hours sleep per day) group, 6hB(6 hours sleep per day and fed fodder enriched with BCAA)group,4h(4 hours sleep per day)group and 4hB group.Re-sults Time of resting on the center lattic,number of make-up and number of excrete were significantly increased in experimental groups compared with C group(P〈0.05),time of resting on the center lattic,number of make-up in 4hB group were lower than that in 4h group (P〈0.05).Total score of open-field test,6h group〉C group〉4h group(P〈0.05).But score of open-field test, number of excrete in 6hB(4hB) group were not significantly different from that in 6h(4h) group(P〉0.05).Conpared with C group,the level of FPG in experimential groups were increased apparently (P〈0.05),and the level of FPG in 4hB group was de-creased markedly compared with 4h group(P〈0.05).The levels of INS were increased significantly in 4h and 4hB groups com-pared with C group(P〈0.05),and 4hB〈4h (P〈0.05).The level of INI in experimental groups were increased apparently com-pared with C group(P〈0.05),and 4hB〉4h (P〈0.05).Conclusions supplementation of BCAA may ameliorates behavior and glucose metabolism in aged rats subjected to isomnia.
