摘    要:建立了毛细管区带电泳法间接测定七种无机阳离子的方法,考察了背景溶液pH值、咪唑和酒石酸浓度等对无机离子分离效果及峰面积的影响。以10mmol/L咪唑-4mmol/L酒石酸(pH=5.5)为背景,在15kV、214nm的条件下,对红细胞消化液中K+、Mg2+、Ca2+和Zn2+进行了定性、定量分析。回收率在96%~104%之间,线性相关系数均在0.9990以上,迁移时间和峰面积的日内相对标准偏差分别低于0.76%和2.83%,日间相对标准偏差分别低于0.85%和3.69%。本方法弥补了原子吸收法不能同时测定细胞内多种离子的不足,有望成为测定细胞内离子含量的检测方法之一。

Abstract:A method for indirect determination of inorganic cations was established by capillary zone electrophoresis with UV detection.The effect of electrolyte composition such as pH in background solution,the concentrations of imidazole and tartrate on the separation and peak area of metal ions was investigated.The concentrations of K+,,Mg2+,Ca2+,Zn2+ in the lysing solution of erythrocytes of the healthy persons were analyzed under the following conditions:background solution of 10 mmol/L imidazole and 4 mmol/L tartrate(pH=5.5);running voltage 15 kV;detection wavelength 214 nm.Under the given conditions the recovery of standard sample ranged from 96% to 104% with good linear relation(r>0.999).The relative standard deviation was <0.76% for migration time and <2.83% for peak area,respectively.Comparing with the atomic absorption spectroscopy the established method can detect several ions simultaneously and is expected to be an useful method for routine analysis of intracellular ion concentrations.
