木犀草素对大鼠肝癌细胞CBRH7919的作用机制

摘    要:从细胞凋亡的角度研究木犀草素对大鼠肝癌细胞CBR.H7919tk/GCV系统的作用机制.采用流式细胞仪技术分析木犀草素砷大鼠肝癌细胞CBRH7919出/GCV自杀基因治疗系统的影响,以及基绿-派诺宁染色、丫啶橙(AO)染色的观察,还用琼脂糖凝胶电泳技术分析了药物对DNA的作用.经过统计学检验:联合组的凋亡率明显高于非联合组;甲基绿-派诺宁染色和丫啶橙(AO)染色出现了较为典型的凋亡形态;琼脂糖凝胶电泳出现明显可辨的梯状条带.综上,木犀草素对CBRH7919墩/GCV系统具有诱导其凋亡的作用.

Abstract:The effects of luteolin on rat hepatoeareinoma cell CBRH7919tk/GCV system from cell apoptosis mechanism is analyzed by using a FACScan cell analyzer to observe the effects of luteolin on CBRH7919tk/GCV suicide gene system. Methyl green-pyronin staining and acri dine orange (AO) staining are used in the cellular apoptotic morphology observation. DNA of the cells is extracted and identified by gel eleetrophoresis. Union group's apoptotie ratios are higher than the non-union group obviously by statistics examination. The result indicated that Methyl green-pyronin staining and acri dine orange staining showed evident apoptotic morphology in luteolin treated CBRH7919 CBRH7919tk/GCV system;that the DNA extracted from the cells showed apparent "DNA ladder". In synthesis, Luteolin can induce the CBRH7919tk/GCV systern cell apoptosis.
