北京地区汉族人群ITGA6基因多态性与前列腺癌的易感性研究

目的探讨北京地区汉族人群中ITGA6基因多态性与前列腺癌遗传易感性的关系。方法采用病例对照研究,提取121例前列腺患者(病例组)和136例非前列腺癌健康人(对照组)外周血中基因组DNA,采用聚合酶链反应-高分辨熔解曲线(PCR-HRM)技术结合测序验证法检测其ITGA6基因rs12621278单核苷酸多态性的分布情况,分析ITGA6基因多态性与前列腺癌的相关性。结果①前列腺癌组中AA,AG,GG基因型分别为69例(57%),40例(33.1%),12例(9.9%);正常对照组中AA,AG,GG基因型分别为69例(50.7%),55例(40.4%),12例(8.8%)。②ITGA6基因rs12621278单核苷酸多态性的三种基因型频率在前列腺癌组和正常对照组间的分布无显著差异(χ2=1.498,P=0.473),两种等位基因频率在两组间的分布也无显著差异(χ2=0.43,P=0.512)。结论 ITGA6基因rs12621278单核苷酸多态性与北京地区汉族人群前列腺癌的发生无明显相关性,可能与中国人前列腺癌发病风险无关。

Association between SNP rs12621278 of ITGA6 gene and risk of prostate cancer in Han population in Beijing

Objective To investigate the correlation between ITGA6 gene polymorphisms and genetic susceptibility to prostate cancer in the Han population in Beijing.Methods A case control experiment was performed,consisting of 121 cases with newly diagnosed prostate cancer and 136 cancer-free healthy controls.Then peripheral blood genome DNA was extracted.Polymerase chain reaction-high resolution melting curve(PCR-HRM) combined with gene sequencing methods were used to determine the distribution of allele and genotype frenquencies of the SNPrs12621278 of ITGA6 gene.The relationship between polymorphism of ITGA6 gene and susceptibility to prostate cancer was assessed.Results In 121 patients with prostate cancer,the AA,AG,GG genotypes were found in 69 cases(57%),40 patients(33.1%),12 cases(9.9%) ;and in 136 healthy controls,the AA,AG,GG genotypes were found in 69 cases(50.7%),55 patients(40.4%),12 patients(8.8%) respectively.There was no significant difference of the distribution of genotype(χ2 = 1.498,P = 0.473) and allele frequency(χ2 = 0.43,P = 0.512) between the two groups.Conclusion There is no obvious association between SNPrs12621278of ITGA6 gene with prostate cancer in Chinese.It may not be the genetic risk factor of prostate cancer.
