摘    要:目的应用选择性环氧合酶-2(COX-2)抑制剂NS-398作用于胰腺癌细胞株PC-3,观察其放射增敏作用,并对其可能增敏机制进行初步探讨。方法体外培养胰腺癌细胞株PC-3;以流式细胞术(FCM)单抗标记技术检测PC-3细胞的COX-2表达水平;以MTT实验检测细胞增殖抑制情况及NS-398对细胞放射敏感性的影响;以FCM分析细胞周期变化,以RT-PCR分析细胞周期相关蛋白P21表达变化。结果 FCM显示PC-3细胞存在COX-2的异常高表达;MTT实验显示NS-398可显著增强PC-3细胞的放射敏感性;FCM显示NS-398联合放射组G0-G1期比率上调;RT-PCR显示NS-398联合放射组细胞内P21mRNA表达上调。结论选择性COX-2抑制剂NS-398可增强胰腺癌细胞PC-3的放射敏感性,其机制可能与上调P21mRNA表达,进而诱导细胞周期阻滞于G0-G1期相关。
