角膜快速冷冻低温保存的实验研究

摘    要:本文用新鲜角膜,在不同浓度抗冻剂中作不同时间的抗冻处理后,采用快连冷冻法进行了低温保存的实验研究,并得到了比较满意的结果.实验中采用了茜素红和台盼蓝联合染色法鉴别内皮活性.实验结果认为,为得到较高的细胞存活率,角膜抗冻处理应尽量减小抗冻剂对细胞的毒性影响,可采用高浓度、短时间的方式;角膜冷却的关键是提高内皮层的冷却速率,可采用双级冷却法.

Abstract:Experimental observation of cryopreservation by means of rapid cooling of pig cornea, which were processed in cryoprotectants, was given in the paper. Combined - staining with alizarin red and trypan blue was used to discriminate the viability of the endothelial cells. Satisfactory results were got and the following conclusion was given: to keep a high percentage of viable endothelial cells, the cryo-processing scheme of high concentation/short time was proved to be effective on reducing the toxicity of cryoprotectants, as the crux of the cooling is to increase the cooling rate of the endothelium, the so called double-stage cooling is preferred.
