SIRT1 mRNA在C2C12成肌细胞增殖过程中的时相性变化

以小鼠骨骼肌成肌细胞（C2C12）为研究对象,用MTT法检测细胞1～5 d内的增殖活性,用荧光定量聚合酶链式反应法检测SIRT1 mRNA表达水平,探讨第Ⅲ类组蛋白去乙酰化酶1（SIRT1）mRNA在C2C12成肌细胞增殖过程中的时相性表达.结果表明,接种后前2天细胞处于潜伏期,3～4 d进入对数增殖期,5 d后逐渐进入平台期；与第1天相比,第2天的SIRT1 mRNA表达没有显著性差异,第3～5天显著性分别增加了1.8、2.3和2.4倍,且SIRT1mRNA表达与细胞增殖活性呈中度正相关,提示SIRT1 mRNA表达与成肌细胞增殖速度密切相关.

SIRT1 mRNA Phasic Expresson During C2C12 Myoblast Proliferation

Mouse skeletal myoblasts（C2C12） as the object of study, cell proliferation activity was assessed by MTT,SIRT1 mRNA expressions were assessed by fluorescence quantitative PCR（RT-PCR）. These findings suggest that the cells were in the incubation period the first two days after transplantation and 3 or 4 days into the logarithmic proliferation phase, and gradually into the plateau after 5 days. Compared with the day 1, there was no significant differences of the SIRT1 mRNA expression in day 2, but differences of the SIRT1 mRNA expression were significantly increased by 1.8-fold, 2.3-fold and 2.4-fold in 3-5 days, respectively. In addition, SIRT1 mRNA expression and cell proliferation activity was moderately positive correlation. These findings suggest that the expression of SIRT1 mRNA are closely related to myoblast proliferation.
