高纯度线粒体DNA的快速提取

摘    要:报道了一种改进的线粒体DNA(mitochondrial DNA,mtDNA) 提取方法,该方法不仅使提取mtDNA的周期缩短了,而且操作简便,提取的线粒体DNA结构完整,并彻底避免了核DNA、线粒体RNA、蛋白质、有机溶剂等的污染;该方法还可以根据不同的试验要求而选取相应的步骤。与其他方法相比较,该方法具有快速、简便、经济、产品纯度高以及mtDNA结构完整等特点,其得率和纯谋匀能满足mtDNA的各种分析要求。

A Rapid and High Purity Method for Isolation of Mithochondrial DNA

Abstract:Mitochondrial DNA is widely used in the studying field of mechanism of disease, heredity and breeding of animals, evolution of creatures, and many valuable results have been obtained. The premise is effective methods of mtDNA isolation and purification. Some methods have been adopted such as density gradient centrifugation, enzyme digestion, high speed centrifugation, column chromatography and Alkaline Lysis. A method is reported in this paper, which can completely avoide the possible pollution by nuclear DNA, mtRNA, protein etc. In isolation and purification of mtDNA, some procedures can be omited according to requirement of experiments. This method is quicker and simpler than other methods, and the product is purer. The quality and purity of mtDNA by this method are suitable to all kinds of studies on mtDNA.
