氧化亚铁硫杆菌的菌数测定法

摘    要:本文根据氧化亚铁硫杆菌(Tkiohacillusferrooxidsans)培养基成分的特性,从多种染色液中筛选出一种草酸铵结晶紫染液,该染液能使菌体染成蓝色,不形成氢氧化铁沉淀,并阐述了不形成沉淀的原因.经统计学方法处理,草酸铵结晶紫涂片染色法应用于氧化亚铁硫杆菌菌数测定,结果准确可靠,较血球计数器法更为快速、简便.除了能测定液相中的菌数,还能测定矿物颗粒表面吸附的细菌.

Abstract:A violet ammonium oxalate crystal dye liquid is selected for thiobacillus ferrooxidans inthe light of the features of the culture medium's ingredients. Using this dye,bacteria can bedyed blue. No iron hydroxide precipitate is formed and the reason is discussed in the paper.The results treated by statistics prove t'hat this method has fine accuracy and reliability and isquicker and simpler than the method using hemacytometer. Besides the determination of bacteria quantities in llquid phase,the same method can also be used to determine the bacteriaquantities adsorbed on the surfaces of ore granules.
